While having a replay system is a nice feature, it is in no ways a necessity for competitive play. In fact, many top gamers at the professional level, i.e. make their living playing games like Starcraft, have advocated that a replay system has diminished the competitive field because so many people can just copy their strategies or see how they play. Starcraft didn't have a replay system when the game first came out either. It was patched in at a later time.

Yeah being able to play all weekend long, Friday - Monday, is a huge deal. Waiting until Tuesday, while the actual release date, seems so far off when we have all weekend off and could be playing. I will not cancel my preorder however.

Eh, I am not sure what the big deal would be if it did not have a replay system. Sure I love watching replays in games like Starcraft, but have you watched a replay of a match of DotA? B o r i n g! I guess I will have to wait and see what the game is like I guess. But generally the more people that are playing the less interesting I find the replay. I prefer watching 1v1 and 2v2 matches.
It would be nice if people who pre-ordered via digital download could begin downloading the game at say 12:00 AM if not earlier, of the release day. Otherwise why not just drive to the store and buy it, it would be faster unless you are on an amazing connection.
I think a lot of people may be uninformed about the depth of each Demigod or the potential for the depth of each Demigod. Demigods have far more depth than they do in a game like Defense of the Ancients (which it sounds like you might be comparing it to). In DotA, each hero is very limited to play strategy in that each hero always gets the same skills, while not necessarily in the same order, ultimately the hero will have every skill maxed.
