Sedna is pretty easy to kill if you are a pure fire build. Open up with fireball, lay down a circle of fire, then nova when she gets close. At level 15, all of these combined will do over 3000 damage if she stays in your circle of fire to keep attacking you.

Towers are not the only way to play a Rook. The fact that you don't know this and yet are telling other people to not play the Rook if they don't know how to play him, is pretty amusing.
In my experience, an instant 1050dmg fireball is more powerful than a spit that does 1500dmg over 10 seconds. During those 10 seconds plenty of things can happen: They can get heals from priests or Sedna. They can drink a potion. They can switch on invulnerability spells/items. They can port back to their crystal. If someone has 1000hp left and they get spat on, they can still survive. If that same person eats a max level fireball they will die.
Well, here's a question. Would you rather have 2500hp and 50% mitigation, or 5000hp and 0% mitigation? The initial connection phase seems a little better for me, but people still drop during the loading screen and I start the game with either one other player, or all AI.

You guys are missing the point. Yes, there are counters to spit. Oak can shield, QoT can shield, sedna can heal, erebus can mist, etc. The purpose of spit is not to take down the enemy DG. It is to put them on the defensive, to annoy the crap out of them, to make them panic and not get as much exp as you are getting out of creeps. Also I notice that most people buy favor items at the very beginning. It's a much better idea to see which DGs you are playing against before making the investment.
The "best" favor item may depend on the map, but it also depends on the demigod. As UB, I would never get anything besides the anklet. The reason is that they are essential in early game when you're harassing enemy DGs with spit. A lot of the time, whether you can get several spits on them (and thus kill them) depends on whether you can catch them as they run. I do it all the time, and it is usually followed by them quitting after they die several times in a row. Could I perform well
